Tiziano Verri is Associate Professor (2006-present; hab. Full Professor in 2017) of Physiology at the University of Salento (formerly University of Lecce), Department of Biological and Environmental Sciences and Technologies (formerly Faculty of Sciences), Lecce, Italy. He got his M.Sc. (Biology) cum laude from the University of Lecce (1989) and Ph.D. (Physiology) from the University of Naples “Federico II”, Naples, Italy (1995). His education/training/appointments include a doctoral fellowship at the Institute of Physiology, University of Zurich-Irchel, Zurich, Switzerland (1993-1994), a post-doctoral fellowship at the Istituto Sperimentale Talassografico “A. Cerruti”, National Research Council, Taranto, Italy (1995-1996), a post-doctoral fellowship at the Laboratory of General Physiology, Department of Biology, University of Lecce (1997-2000), and the post of Assistant Professorship (Physiology) at the University of Lecce (2000-2006). As an academic visitor/research scientist, he was at the Department of Biology, University of Padua, Padua, Italy (1995-1996), the Institute of Nutritional Sciences, University of Giessen, Giessen, Germany (1998), the Department of Zoology, University of Hawaii at Manoa, Honolulu, Hawaii (1999), the Department of Biology, University of North Florida, Jacksonville, Florida (2002), the Institute of Nutritional Sciences, Technical University of Munich, Freising, Germany (2004). His major research interests cover human physiology, comparative physiology, animal model physiology, epithelial physiology, biophysics, functional genomics and bioinformatics.

This course aims at providing students with an in-depth knowledge of the current view of membrane biophysics, the role as a barrier played by the membrane and the roles played by channels, carriers and receptors in the membrane physiology. The course also aims at highlighting the spatial organization of membrane proteins, their structures and how their major structural elements make them work efficiently. Major methodological approaches to membrane biophysics including their powers and limitations will be also discussed.

Learning methods consist of formal lectures and integrative lectures held using slides and hypertext links to specific Web sites (5 credits = 40 hours), followed by practical laboratory classes (1 credit = 10 hours). Outside these activities, the students are expected to read assigned papers from the scientific literature.

Oral examination. It is aimed at ascertaining, in proportion:

- the level of theoretical knowledge through the presentation of the program topics (50%)

- the level of practical abilities through description of methods and methodologies (20%)

Conoscenze di base di Fisiologia Generale

Il corso si propone di fornire conoscenze relative alla fisiologia degli organismi acquatici (pesci e crostacei) con particolare riferimento a specie di interesse per l’acquicoltura trattando in maniera approfondita gli aspetti relativi:

- all’ omeostasi osmotica, ionica ed energetica in relazione agli ambienti di allevamento ed alle possibili applicazioni pratiche;

 - alla fisiologia della riproduzione e possibili applicazioni pratiche;

 - alla fisiologia dello stress negli ambienti di allevamento.

Acquisizione di conoscenze relative alla fisiologia degli organismi acquatici (pesci e crostacei) con particolare riferimento a specie di interesse per l’acquicoltura. In particolare saranno trattati in modo approfondito: a) i processi di omeostasi osmotica, ionica ed energetica in relazione agli ambienti di allevamento ed alle possibili applicazioni pratiche; b) la fisiologia della riproduzione; c) fisiologia dello stress.

Le conoscenze e le competenze relative agli obiettivi saranno acquisite attraverso lezioni frontali in aula e studio guidato.

La capacità di applicare le conoscenze sarà acquisita attraverso esercitazioni di laboratorio.

Lo studente acquisirà, inoltre, mediante le esercitazioni di gruppo, la capacità di: agire, comunicare e valutare

Modalità di erogazione: Tradizionale Organizzazione della didattica: 5 CFU (48 ore) di lezioni frontali + 1 CFU (12 ore) di esercitazioni di laboratorio. La didattica frontale sarà supportata da proiezione in aula di diapositive e filmati. I file .pdf delle lezioni saranno disponibili sul sito docente

Prova orale mirata ad accertare, in misura proporzionale:

- acquisizione delle conoscenze e competenze previste dal programma (80%);

Il Corso si propone di approfondire il funzionamento dell’organismo umano mettendone in evidenza i rapporti struttura-funzione. Si propone di definire il concetto di omeostasi e di fornire, in particolare, conoscenze sulla funzione endocrina, cardio-circolatoria, respiratoria, renale e gastro-intestinale. Pur non essendo previste propedeuticità, si ritiene importante che lo studente possieda conoscenze di base di fisiologia generale, biochimica e anatomia dell’uomo.

La modalità di erogazione della didattica è del tipo tradizionale, con 8 CFU (64 ore) di lezioni frontali in aula e 1 CFU (10 ore) di attività di laboratorio. Le lezioni in aula prevedono l’utilizzo di diapositive, talora con collegamenti ipertestuali a specifiche pagine Web.

La valutazione degli studenti è effettuata mediante prova orale. E’ mirata ad accertare, in misura proporzionale:

- Il livello delle conoscenze teoriche acquisite, attraverso la presentazione di argomenti del programma (50%)

Conoscenze introduttive dei principi di biochimica, genetica, biologia molecolare e fisiologia

Argomenti di lezione ed esercitazione.

Il corso si propone di introdurre lo studente ai principali problemi classici della bioinformatica come la gestione e la consultazione delle banche dati, l'allineamento di sequenze, l'evoluzione molecolare, l'identificazione e la ricerca di pattern funzionali e la predizione della struttura proteica. Il corso mira a far conoscere allo studente le potenzialità dei moderni sistemi informatici e del web a supporto dei problemi biologici.

This course deals with the basic theoretical physics underlying biological phenomena, with emphasis on those occurring at the membrane level. From the broad axiom that in biological systems ‘a barrier separates two-compartments’ it focuses on the processes that allow a particle (ion, small molecule, protein, nanoparticle, …) to cross the membrane and on how it does interact with water (solvent) and soluble components (solutes) in each compartment. It also focuses on those biophysical phenomena that make a membrane ‘active’, i.e. that allow a biological membrane to produce and transmit a signal from a compartment to the other and/or along the barrier. In this respect, it explores the molecular foundations of biophysics, the thermodynamics of non-equilibrium systems and membrane transport, with emphasis on nerve impulses, and membrane transduction, with emphasis on membrane receptor-based processes.

The course begins with an overview of molecular biophysics and the concept of molecular recognition, followed by a discussion of the interaction between molecules, the primary process that determines membrane transport and reception, and the importance of intercellular interactions in the existence and development of multicellular organisms.

The student is also introduced to the stability conditions of a steady-state, the concept of entropy for an open system, the thermodynamics of the sodium-potassium pump, ionic equilibrium between sodium- and potassium-rich solutions separated by an active membrane, the conformational properties of membranes, and the general phenomenological theory of active, facilitated and passive transport and the role of the carriers and channels.

The structure-function relationships that allow channels and carriers, on one side, and receptors, on the other, to operate efficaciously in the membrane are discussed on the basis of the most recent structural data available in protein databanks.

This course is a valuable resource for biologists and biotechnologists, physicists and biophysicists, graduate and postgraduate students having basic knowledge of physics, and anyone acquainted with biological membranes and proteins.

The student is also introduced to the stability conditions of a steady-state, the concept of entropy for an open system, the thermodynamics of the sodium-potassium pump, ionic equilibrium between sodium- and potassium-rich solutions separated by an active membrane, the conformational properties of membranes, and the general phenomenological theory of active, facilitated and passive transport and the role of the carriers and channels.

The structure-function relationships that allow channels and carriers, on one side, and receptors, on the other, to operate efficaciously in the membrane are discussed on the basis of the most recent structural data available in protein databanks.

This course is a valuable resource for biologists and biotechnologists, physicists and biophysicists, graduate and postgraduate students having basic knowledge of physics, and anyone acquainted with biological membranes and proteins.

La valutazione degli studenti è effettuata mediante prova orale. E’ mirata ad accertare, in misura proporzionale:

- Il livello delle conoscenze teoriche acquisite, attraverso la presentazione di argomenti del programma (50%)

- Il livello delle abilità pratiche acquisite, attraverso la descrizione di metodiche e metodologie (20%)

- La capacità di applicare le conoscenze teoriche e le abilità pratiche acquisite alla soluzione di problemi semplici (30%)

No formal prerequisites are required with respect to other courses. However, basic knowledge of general physics, physical chemistry and general physiology is recommended.

This course deals with the basic theoretical physics underlying biological phenomena, with emphasis on those occurring at the membrane level. From the broad axiom that in biological systems ‘a barrier separates two-compartments’ it focuses on the processes that allow a particle (ion, small molecule, protein, nanoparticle, …) to cross the membrane and on how it does interact with water (solvent) and soluble components (solutes) in each compartment. -           Principal investigations (basic science)

Research activity is mainly centered on the examination of the transport processes/mechanisms that mediate the flux of solutes across epithelia. Major research interests cover the function and regulation of those SoLute Carrier (SLC) membrane transport proteins that mediate the flux of small solutes (sugars, amino acids, vitamins, peptides, peptido-mimetics and peptide-like drugs) across the plasma and organelle membranes of (epithelial and non-epithelial) cells. To support this research, the project proponent routinely applies physiological and biochemical methodologies, as well as molecular biology and bioinformatics, and he uses both in vitro systems (cell lines, Xenopus laevis oocytes) and animal models, either conventional (mammals) or alternative (teleost fish and crustaceans). Among the latter is the cyprinid Danio rerio (zebrafish). Major published results regard the studies on the function and regulation of members of the SLC15 family of the H+-oligopeptide cotransporters, e.g. SLC15A1(PEPT1), SLC15A2(PEPT2), SLC15A4(PHT1), and SLC15A3(PHT2), of the SLC13 family of the Na+-sulfate/carboxylate cotransporters, e.g. SLC13A1/NaSi, and of the SLC34 family of the Type II Na+-phosphate cotransporters, e.g. SLC34A1/NaPi-2a, in animal and cellular models.

-           Principal investigations (applied science)

Early experimental activity has been centered on the development of new strategies to improve gene transfer technology. Part of this applicative/biotechnological research regarded the somatic expression of heterologous genes in teleost fish and the analysis of the related processes of transcriptional activation, and it has also been conducted using both adenoviral vectors specifically designed for gene therapy and vectors based on transposable elements as gene vehicles. An aspect of this applicative/biotechnological research has specifically regarded the setup of protocols for DNA vaccination in teleost fish; this research on fish vaccines is still ongoing.

After his first enrollment on a permanent position at the University of Lecce (2000), Tiziano Verri has definitely focused on the physiology of digestive tract of lower vertebrates, with special emphasis to fish nutritional physiology, diet formulations, fish health, and human food production. Part of his research has been devoted to understand how efficiently amino acids as single molecules and/or in the form of small peptides (di- and tripeptides) are absorbed, metabolized and utilized for fish growth. These results have found a natural application in aquatic laboratory (zebrafish) and aquaculture (salmon and salmonids, cod, carp, sea bream, etc.) animal nutrition, health and welfare. At the moment, his collaborative network on fish digestive physiology and nutrition includes (to mention): prof. In more recent years (2012-to date), functional genomics approaches have been developed and used to support/complement researches in human biology and pathology (with emphasis on rare disease). In particular, structure-to-function analyses have been used, based on the assumption that the evolution of vertebrates has been accompanied by the evolution of their own parts at each different level of biological organization. Thus, comparing information contained in (ortologous-paralogous) biosequences (e.g. nucleic acids, proteins, small peptides, etc.) from human to those obtained from very distantly related species along the vertebrate scale (fish included) can represent a valuable tool to analyze “a priori” protein evolution. Combined to the study of the whole set of missense variants in a protein, this comparative approach has proven to be very useful in solving the question of the role of proteins (e.g. sacsin) unknown for function but known for being involved in rare genetic diseases. This translational approach can virtually be applied to the study of every single gene/group of genes of interest, with the possibility to identify stretches/domains/regions/etc. unknown but anyhow “conserved” and possibly relevant for playing a role in a pathology. At the moment, his collaborative network on functional comparative genomics of rare disease includes (to mention): dr.
